Isolation and identification of the infectious haematopoietic necrosis virus (IHNV)-like agent from farmed rainbow trout (Oncorhynchus mykiss) from Iran.

Abstract

The isolation of viral causative agent of fry trout mortality syndrome in farmed rainbow trout in Iran is presented. A total of 420 samples were collected from six provinces (October 2001 to May 2002) for virological, serological and electron microscopic examinations. No cytopathic effect (CPE) was observed in all the cell cultures except for EPC and BF-2 cells in which signs of CPE appeared one day postinoculation during the second and third passages of two brooder ovarian fluids. Electron microscopic examination revealed viral particles measuring 67-73Ă—128-182 mm in the pelleted materials from the CPE-positive BF-2 and EPC cell lines. Indirect immunofluorescence antibody test were positive for samples collected from the kidneys of the larvae. Ovarian fluid samples showed CPE on EPC and BF-2 cell lines as well as CPE positive cell cultures. Using ELISA, eight of all serum samples tested were positive with optical density ranging from 0.316 to 2.268 compared to 0.06 for negative controls.
