Nucleic acid spectrophotometric analysis for the genetic characterization of some Chinese carp species habituated in Romania.

Abstract

Muscle samples were removed from 2 silver carp (Hypophthalmichthys molitrix), 2 bighead carp (Aristichthys nobilis) and 2 grass carp (Ctenopharyngodon idella), 2 years of age. DNA was extracted for karyotype analysis and the nucleic acid content was analysed by absorption spectrophotometry at 230-290 nm. The 3 species all had the same chromosome number (2n = 48) and similar chromosome morphologies and dimensions. Spectrophotometric analysis revealed similar average values of the total nucleic acid content in the cells of the 3 species (0.0265, 0.0263 and 0.0255 mg/ml tissue for H. molitrix, A. nobilis and C.idella respectively). It is concluded that the 3 species are closely related but that the chromosomes of C. idella contain slightly less chromatin than those of the other species.
