Abstract

Details are given of the protocol followed in preparing a clone of peanut stunt cucumovirus RNA4, the production of single-stranded DNA templates of both strands of PSV cDNA and sequencing using progressive oligonucleotide primers. The nucleotide sequence of the cDNA clone is figured, beginning with the 5′-terminal nucleotide and including an open reading frame (ORF) of 678 bp (including the stop codon), a 5′ noncoding sequence of 46 bp and a 3′ untranslated sequence of 262 bp. The ORF encodes a PSV capsid protein of 224 amino acids with molecular mass of 24.9 kDa.
