Abstract

Micropropagation of Acacia mangium Willd. through nodal bud culture.

Abstract

In vitro multiple shoot proliferation was achieved in A. mangium using nodal explants of an 8-yr-old elite tree. The maximum number of shoots were obtained on MS medium supplemented with BA ([benzyladenine] 3 mg/litre) and NAA (0.1 mg/litre). Addition of ascorbic acid (100 mg/litre) to the shoot proliferation medium inhibited browning of the medium. Shoots regenerated in vitro were rooted on MS medium supplemented with IBA (1 mg/litre) and IAA (0.5 mg/litre). Genotypically uniform regenerated plantlets survived (70%) in soil after hardening.
